Abstract

Tissue culture is a method of propagation by taking cells or tissues or plant organs that are carried out aseptically so that new plants or individuals can be obtained. The use of in vitro techniques for the purpose of vegetative propagation is the most advanced technique in tissue culture. The provision of potatoes in Indonesia still cannot meet the needs of the community. This is due to the difficulty of procuring potatoes in large quantities and in a short time. In response to the many problems that occur, one of the efforts that can be done is to propagate potato plants in vitro or tissue culture by using plant growth regulators (ZPT). The tools used in tissue culture techniques are LAF, oven, autoclave, hot plate and stirr, pH meter, freezer, analytical scales, measuring tubes, beaker glass, volume flasks, petri dishes, media bottles of various sizes, volumetric pipettes, erlemeyer tubes, tweezers, sterile knives, sterile scissors, media tubs and push baskets. The materials used in this study are 70% and 96% alcohol, distilled water, methylated spirits, Bunsen burner, matches, sterile distilled water, sterile filter paper, aluminum foil, seal, cotton, ½ K0.5 media as control, ½ K0.5 + 2% mannitol as treatment, potato explants (Solanum tuberosum L.). The technique of propagating potato plants in vitro tissue culture can be done by selecting tools and materials, washing and sterilizing the media, washing and sterilizing the media, making stock solutions and potato culture media, giving Growth Regulators (ZPT), preparing explants, planting explants, maintaining the culture. In the process of plant propagation with in vitro tissue culture techniques should be done carefully. Make sure all tools, materials and media used are clean and sterile so as not to contaminate the explant plants to be cultivated.
